Paraformaldehyde-fixed, paraffin embedded human breast carcinoma; Antigen retrieval by citrate buffer (pH 6.0 ), boiling bathing for 15min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30min; Blocking buffer (normal goat serum) at 37℃ for 20 min; Antibody incubation with anti-WNT6 antibody (Cat#52644, 1:200), overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining.
Western blotting analysis using anti-WNT6 antibody (Cat#52644). Total cell lysates (30 μg) from various cell lines were loaded and separated by SDS-PAGE.The blot was incubated with anti-WNT6 antibody (Cat#52644, 1:1,000) and IRDye800CW-conjugated goat anti-rabbit secondary antibody (1:20,000) respectively.
